Long-Read High-Throughput Sequencing (HTS) Revealed That the Sf-Rhabdovirus X+ Genome Contains a 3.7 kb Internal Duplication

We previously reported a novel rhabdovirus produced from the Spodoptera frugiperda Sf9 cell line, designated as Sf-rhabdovirus X+ since it contained unique accessory gene X. The genome sequence was generated using Sanger sequencing and short-read high-throughput (HTS). In this study, we have used long-read HTS technologies, PacBio’s single-molecule real-time Oxford’s Nanopore RNA direct sequencing, to analyze parent line transcriptome virus an clone, respectively. A 3.7 kb duplication identified in L between nucleotide position 8523 8524, preceded by GA dinucleotide insertion. This partial G gene, complete X which extended positions 4767–8523 virus. Thus, length is 17,361 nucleotides, re-designated X+3.7. found all clones producing variant Furthermore, X+3.7 stable at passage 30, highest tested. These results highlight importance of combining technologies for accurately genomes HTS.
